Hexokinase specificity of some tumor tissue extracts.

چکیده
The substrate specificity of hexokinase preparations from Ehrlich ascites cells, a spontaneous adenocarcinoma, Jensen sarcoma, and Walker carcinoma was studied with a number of sugars which differed in structure from glucose at one or more carbon atoms used as substrates. A marked variat ion in specificity was found. The preparations from Jensen sarcoma and Ehrlich ascites cells were relatively specific for glucose and some C-~ analogs. The preparations from the adenocarcinoma and Walker carcinoma were nonspecific and brought about phosphorylat ion of a number of sugars differing from glucose in many positions. Inhibit ion of the hexokinase preparat ion from Ehrlich ascites tumor cells by altrose and talose was observed.

Spectrophotometric measurement of hexokinase and phosphohexokinase activity.
The methods available for the quantitative assay of these enzymes are based on phosphate transfer determinations and pH change of the reaction mixture. They require large quantities of adenosine triphosphate and, for estimation in crude tissue extracts, the addition of inhibitors such as iodoacetate and sodium fluoride (7). The purpose of this paper is to report a simple and rapid spectrophotom...
متن کاملThe hexokinase reaction in tissue extracts from normal and diabetic rats.
The recent papers by Cori and his coworkers have stimulated interest in the hexokinase reaction in tissue extracts from normal and pathological animals. Our interpretation of this work pertinent to the subject of this paper is as follows: (1) Rats injected with alloxan yield muscle extracts which show an initial lag of hexokinase activity of 10 to 15 minutes duration, owing t,o the presence in ...
